【摘要】:ThistestmethodhasbeendevelopedinsupportoftheNationalHomelandSecurityResearchCenter,USEPAbyRegion5ChicagoRegionalLaboratory(CRL).Bromadiolone,brodifacoum,diphacinoneandwarfarinarerodenticidesforcontrollingmice,rats,andotherrodentsthatposeathreattopublichealth,criticalhabitats,nativeplantsandanimals,crops,foodandwatersupplies.Theserodenticidesalsopresenthumanandenvironmentalsafetyconcerns.Warfarinanddiphacinonearefirst-generationanticoagulants,whilebromadioloneandbrodifacoumaresecond-generation.Theanticoagulantsinterferewithbloodclotting,anddeathcanresultfromexcessivebleeding.Thesecond-generationanticoagulantsareespeciallyhazardousforseveralreasons.Theyarehighlytoxicandpersistalongtimeinbodytissues.Thesecond-generationanticoagulantsaredesignedtobetoxicinasinglefeeding,buttime-to-deathoccursinseveraldays.Thisallowsrodentstofeedmultipletimesbeforedeath,leadingtocarcassescontainingresiduesthatmaybemanytimesthelethaldose.Thismethodhasbeeninvestigatedforusewithreagent,surface,anddrinkingwaterfortheselectedrodenticides.1.1Thisprocedurecoversthedeterminationofbromadiolone,brodifacoum,diphacinoneandwarfarin(referredtocollectivelyasrodenticidesinthistestmethod)inwaterbydirectinjectionusingliquidchromatography(LC)anddetectedwithtandemmassspectrometry(MS/MS).Theseanalytesarequalitativelyandquantitativelydeterminedbythismethod.Thismethodadherestomultiplereactionmonitoring(MRM)massspectrometry.1.2TheDetectionVerificationLevel(DVL)andReportingRangefortherodenticidesarelistedinTable1.1.2.1TheDVLisrequiredtobeataconcentrationatleast3timesbelowtheReportingLimit(RL)andhaveasignal/noiseratiogreaterthan3:1.Fig.1displaysthesignal/noiseratiosoftheprimarysinglereactionmonitoring(SRM)transitions,andFig.2displaystheconfirmatorySRMtransitionsattheDVLsfortherodenticides.1.2.2ThereportinglimitwascalculatedfromtheconcentrationoftheLevel1calibrationstandard,asshowninTable4,accountingforthedilutionofa40mLwatersampleuptoafinalvolumeof50mLwithmethanoltoensureanalytesolubility.1.3Units8212;ThevaluesstatedinSIunitsaretoberegardedasstandard.Nootherunitsofmeasurementareincludedinthisstandard.1.4Thisstandarddoesnotpurporttoaddressallofthesafetyconcerns,ifany,associatedwithitsuse.Itistheresponsibilityoftheuserofthisstandardtoestablishappropriatesafetyandhealthpracticesanddeterminetheapplicabilityofregulatorylimitationspriortouse.
