【摘要】:Byfollowingthegeneralguidelines(Section5)andinstrumentcalibrationmethods(Sections6-16)inthisguide,usersshouldbeabletomoreeasilyconformtogoodlaboratoryandmanufacturingpractices(GXP)andcomplywithregulatoryandQA/QCrequirements,relatedtofluorescencemeasurements.Eachinstrumentparameterneedingcalibration(forexample,wavelength,spectralresponsivity)istreatedinaseparatesection.Alistofdifferentcalibrationmethodsisgivenforeachinstrumentparameterwithabriefusageprocedure.Precautions,achievableprecisionandaccuracy,andotherusefulinformationarealsogivenforeachmethodtoallowuserstomakeamoreinformeddecisionastowhichmethodisthebestchoicefortheircalibrationneeds.Additionaldetailsforeachmethodcanbefoundinthereferencesgiven.1.1Thisguide(1)liststheavailablematerialsandmethodsforeachtypeofcalibrationorcorrectionforfluorescenceinstruments(spectralemissioncorrection,wavelengthaccuracy,andsoforth)withageneraldescription,thelevelofquality,precisionandaccuracyattainable,limitations,andusefulreferencesgivenforeachentry.1.2Thelistedmaterialsandmethodsareintendedforthequalificationoffluorometersaspartofcomplyingwithregulatoryandotherqualityassurance/qualitycontrol(QA/QC)requirements.1.3Precisionandaccuracyoruncertaintyaregivenata1x03C3;confidencelevelandareapproximatedincaseswherethesevalueshavenotbeenwellestablished.1.4ThevaluesstatedinSIunitsaretoberegardedasstandard.Nootherunitsofmeasurementareincludedinthisstandard.1.5Thisstandarddoesnotpurporttoaddressallofthesafetyconcerns,ifany,associatedwithitsuse.Itistheresponsibilityoftheuserofthisstandardtoestablishappropriatesafetyandhealthpracticesanddeterminetheapplicabilityofregulatorylimitationspriortouse.
